Studies on Insulin-dependent diabetes in STZ-induced SHR


Abstract
This study was carried out to investigate the relationship between hypertension, diabetic nephropathy and changes of pancreas. The 8-week-old spontaneously hypertensive rats(SHR) with insulin-dependent diabetes mellitus were induced by streptozotocin(STZ, 60§·/§¸ body weight) treatment intraperitoneally. The body weight; blood glucose, urine glucose and urine ketone body were measured every week. The blood pressure and heart rate were measured every 2 weeks. Light microscopical studies were carried out in both pancreas and kidney. Body weight was decreased in STZ-SHR after injection of STZ. Blood glucose and urine glucose were increased in STZ-SHR. Urine ketone body was increased for the first 2 weeks. Thereafter it was decreased. The systolic and diastolic blood pressure, heart rate of STZ-SHR were decreased than those of SIR rats, By contrast, the systolic and diastolic blood pressure of SHR rose with aging. STZ-SHR showed necrosis and vacuolations in pancreatic islets. And STZ-SHR showed glomerutar hypertrophy, increasing of mesangium and fibrosis in glomerulus, gaining of kidney weight. These observations suggest that the renal diabetic changes were distinct in STZ-SHR. And decreasing blood pressure possibly due to insulin deficiency and morphological changes of heart in STZ-SHR.
